Hi Gibbsy. Is there any data on this drive?
My USB external drives will not show up unless they are formatted with a drive letter.

Try USB2 (black port) reboot, and see is anything shows.

To add to Dougie’s suggestion is there another pc you can try it on?

              and something else to try.
it may be the external caddy itself.
try putting the drive directly into the PC.

and a cautionary note on external drives. (not saying this applies to you - just a heads up to all out there )
all drives hate vibrations, but generally the surrounding cases of laptops and desktops offer a bit of a buffer zone.
              not so with external enclosures.
my analogy is, if the force is enough to crack an egg, it’ll be enough to destroy data or the drive itself.
so don’t keep them within arms reach when they are in use and if you have to move them, disconnect first, then move. (or be bloody careful)
when they are off (disconnected) they are almost indestructible.

Only yesterday, I had an SSD in an external enclosure, and would not show up in Disk Management,Device Manager.
Switched from USB3 to USB2 rebooted, and showed up in Disk Management, and Device Manager.
The USB3 ports are working as I use it for my Flash drives, webcam etc,
The external drive just would not be recognized in the USB3 ports.
                How weird is that?

as Dougie said, USB2 ports usually have a black bit of plastic inside that forces you to orientate the USB plug only one way.
                        USB3 are usually blue.

but in both cases - not always.

if it also fails on other PC’s, it’s looking more like a caddy or drive itself issue.

Would putting the HDD in to the second internal slot help, or just the same result?

that would definitely help, and in fact is the next logic thing to try, if for no other reason then to prove it is the drive and not the enclosure.
if drive is detected, sweet - it’s the caddy.
if drive still not accessible, β€˜pile of poo emoji’ - you’re in a world of hurt.
